I'm probably wrong, but I'd play it like this:

e|--4--|
b|--4--|
g|--5--|
d|--6--|
a|--6--|
e|--0--|

The chord labeled G#/E in the tab is actually E/G#. I presume E/G# is what he's after.

yea that's an E/G#

I would play it this way:

E||--0--||
B||--0--||
G||--1--|| <--- 1st finger
D||--2--|| <--- 2nd finger
A||-----||
E||--4--|| <--- 4th finger (AKA pinky loll)
you'll have to either mute the A string (with your pinky or 3rd finger) or play it with your fingers instead of a pick... cause the A string shouldn't be heard on this chord!

Actually, you could play it this way too, it would be much simpler!

E||--0--||
B||--0--||
G||--1--|| <--- 1st finger
D||--2--|| <--- 2nd or 3rd finger
A||--2--|| <--- 2nd or 3rd finger
E||--4--|| <--- 4th finger (AKA pinky loll)
for the A and D strings, just use whichever finger you feel comfortable with... but use both fingers (1 per string) cause if you barre it, well it's gonna be painful for nothing...

Or if you're lazy:

E||--0--||
B||--0--||
G||--1--||
D||-----||
A||-----||
E||-----||

Assuming that we are indeed looking for an E/G# (a.k.a. an E chord in first inversion) instead of G#/E (highly unusual):

here's a power chord version:

-x-
-x-
-x-
-x-
-7-
-4-

here's a common triad version. Mute first and second strings with first finger

-x-
-x-
-4- pinky
-2- first finger
-x-
-4- ring (third) finger

... or leave them open

-0-
-0-
-4- pinky
-2- first finger
-x-
-4- ring (third) finger

here's another shape, this'll get your fret hand stretched. Used in Crash by Dave Matthews band:

-x- or open
-5- pinky
-4- ring (third) finger
-2- first finger
-x-
-4- second finger

It's not neccessary to play the third string first fret (G#) if you're fretting it in the bass (sixth string fourth fret), indeed its counterproductive First inversion chords (third in the bass instead of the root) are more commonly voiced with just root and fifths above the third in the bass
