If I play the 5th fret on the B string and an open E, I can tune that interval perfectly. However, when I play an open G string, and third fret on the high e (an octave), my E string is about 20-30...

Breathe and push from your diaphragm. Really. That will get you 75% of the way there. Make sure you can do this properly though. Takes a TON of practice not to ruin your voice and get a good tone
