some strings just sound flat all the time especially as they age.
if you do swap strings, go for a similar gauge and swap one at a time.

you can tune it up. if it's set up correctly, the bridge should still be balanced and their shouldn't be buzz transmitting trough the amp.

In this case I would say, no, don't do it.
