I wouldn't bother. You can get tube amp kits for guitar, I'd do that. A headphone amp is going to give you trouble with output, impedance, and linearity/gain structure that will probably make it unpleasant sounding from a guitar perspective.

It wasnt something I was going to attempt but I was just curious as to if it would work or evern be possible

As they are really cheap compared to a regular guitar amp pcb

Yeah...there's a reason for that. A single 12AU7 tube putting out a fraction of a watt of squeaky clean linear volume isn't exactly guitar territory. It will work with a guitar, but so would plugging into a telephone. Both are likely to sound like total crap.

Start out with the noisy cricket or the ruby amp build, those are small and cheap and actually sound decent with a guitar. If you really have to have a tube I'm sure there are similar builds out there that have one, but

The noisy cricket does look good but i'm no amp builder, and if I were i'd have to go with something that has 3 band eq
