I have a Fulltone Fulldrive 2 Mosfet and I can't seem to get THAT tone. I mean it never gives me a bad tone, they're always great coming out of that little blue box. I just can't get the sound I want. I run it into a Blues Junior. I'm looking for a tone like this

Especially those first few bends, great. So, anyone with some experience would be appreciated.

Use the neck pup, set all the controls at noon on the pedal and, set it to FM or vintage mode, use the mosfet and turn the boost on. That'll probably be close.

What kind of tubes are in the Blues Jr.?
Getting some nicer ones might smooth out your sound.
Also play with your volume and tone knobs lots, and work on your vibrato technique.

That's actually a pretty good point, the stock GTs (rebranded sovteks) are pretty harsh, I run a JJ and a mullard in mine, but just jjs would be good.

Id' rather say a new speaker...

For settings, use the vintage mode. Then set the volume high and adjust the gain, don't use the boost.
