If I blocked off the floyd rose trem on a guitar, would i have to adjust it when changing tunings?

If you blocked it so that it can't move in either direction, then no you won't have to adjust to change tunings, although you might need to tweak the intonation.

Why would the intonation need tweaking?
If the trem is blocked then it wont move :P

unless you mean it'll need tweaking once for it's blocked position

He wanted to know if he would need to make any adjustments after changing tunings. When you change tuning you change the tension on the strings, which can affect the intonation.
