yes, but not in the way i think you are thinking. with an effects loop you typically have two long cables to/from the amp. if all your pedals are true bypass, you are going to lose some highs. i would say you want at least one buffer in there.

what wouldnt negatively affect your tone?

also, there are so many myths about true bypass. it isnt as great as people make it out to be. or as bad as others claim. worry more about what the pedal sounds like while on.
