Does putting a blanket/matt under your amp while playing live really make you sound better?
If so, how and why? Would it benefit me to do it?

I could maybe see a purpose if your amp had casters that didn't lock and it was on something like a gym floor. But as far as sound, no. If it did anything, I can't imagine it would be good.

put a blanket over your cab.

play like that for a few days.

take it off.

it will sound better.

It depends, sometimes it can help a bit on a hard floor - stops things being so boomy. But seriously you'll look stupid turning up at a gig and rolling out a blanket to place your amp on.

An amp stand on the other hand is a different story.
