You loosen the nut blocks (near the tuners) this allows you to us the tunners.
When you get it in tune you use the hex key and lock the nut back down (don't over tighten them)
Then you use the fine tuners (on the back of the bridge) to get it set perfect. I have found it is easiest to have the fine tuners in the middle of the range of motion.

When you change strings only do I at a time.

It has a Floyd Rose tremolo so has 2 different tuners. The tuners on the headstock that you're using that don't work are locked by the nut where the neck meets the headstock. To use those you need to loosen the lock with an Allen key before you can use them.

At the opposite end you have fine tuners which aren't locked. Turn them clockwise to tune up and anti-clockwise to tune down.

You'll need to keep tuning and re-tuning several times before you get it sorted.
