Overdrive makes your guitar sound distorted to some effect. Overdrive has a softer tone than distortion and fuzz.

Gain is how much overdrive, distortion, or fuzz you have.

gain is the technical term between the power of a signal and the power of a signal after it's amplified. overdrive is when you give your amp too much gain.

Overdrive is basically raising the volume of the signal and overdriving it so it sounds dirty. Gain is how much overdrive is being used.

If you're wondering what distortion is (I can't tell from the way you asked your question) and you're wondering the difference between overdrive and distortion, distortion is an effect that raises the wavelengths of your signal and makes is bigger, thus adding distortion.
