Would you be able to run straight from a Boss Gt-6 to a speaker cabinet or would the sound be affected by not running it through an amp?

you'd pretty much get no sound. Preamps don't generate enough power to drive a speaker.

i don't know what the gt-6 is but from dirk gently's post, i can assume it's a guitar preamp...if you wanted to do that, you'd need to get a power amp, because an amp is made up of 3 things:

these are the general definitions

1) preamp gives [sound]
2) power amp [gives power]
3) cabinet [speakers, amplifies sound]

you can bypass the preamp completely my connecting into the efx loop return or CD input (if you amp has either of those)
