I have just bought a BOSS TU-15 chromatic tuner and I am going to do the intonation on my Les Paul.

I'm not quite clear on one thing though: I know I should use new strings to do the intonation, but should I do it immediately after I put the strings on and stretch them? Or should I play it for a bit and then do the intonation after the strings have been used for an hour or so?
