You can, I have the three channal programmable one. My rig is kept at a friends house for band practice, and ill just run it through the loop. Ill also take the pedal home ane just run in the signal chain for a small crappy practice amp I have. It really makes the practice amp sound a lot better, it fattens everything up and makes it warmer. Great pedal.

I just looked at the booklet for mine and it says run the output to the effects return, so i guess the mean effect loop, but they worded it very badly, I know it works both ways though.

Does this mean using it as a preamp and bypassing the amp's built in preamp? Kind of what it sounds like to me

No, that means to use it as your preamp, and ignore the one of your amp, hence, going to the effects return.
