Uhm.. well I can't really offer a lot of help, but doesn't he use Marshall Silver Jubilee amps? Or is that his stage gear, because those have a very distinct sound (I love them)

I'd say his basic tone would be:

Black 2x12 model, the volume cranked, the gain at about 9 o'clock. Treble, noon. Mid, 3 o'clock. Bass, 2 o'clock. Maybe add some reverb set at about 10 o'clock.
(Using neck and middle pickups.)

Now, this is all off the top of my head. I have a Valvetronix and ****ing love it. I was messing around and twisting knobs and got pretty close to his tone, and I think it was about that.
Go off that and tweak it a bit. You'll find it.

I've heard it's not easy getting his tone without some slightly over-wound formvar pick-ups I believe.

try spending several grand on a dumble amp. (he uses it in the studio.)
or any american sounding clean amp, neck and middle pickups. slightly (very slightly) roll back the mids.

Actually guys he uses a Fender Blues Junior in the studio, (if you don't believe me, check out this: http://www.johnmayer.com/blog). He also uses Two Rock amps on stage.

he used that like once, not even for the record, just playing around with "red house"

interestingly if you look thru the liners on the Continuum CD inset you will see a ton of his gear porn.

i didn't think he used a BJ either but I have heard that he used the Two Rocks

On all the studio programs i've seen him, he's had his two rocks.

And he goes for a classic blackface, 60s fender formvar slightly overwound pickup tone.

So you'll want a fair bit of bass, neck+middle, scooped mids, glistening treble.
The Laney Thread are big and clever. No exceptions.

i just remember hearing in an interview that his prized dumbles are his "secret weapon" in the studio. Two Rocks are basicaly dumble clones though.

and when i say studio, i mean when hes recording albums. i still see his dumble ods on stage occasionaly.

Either the Boutique Clean model or the Black 2x12 model, scooped mids, 2/3 bass, 5/6's treble, neck or middle pickup, tone knob rolled back to taste.

I personally hate his tone because he scoops the **** out of it.

He doesn't scoop a lot out of it, he has formvar pickups and a blackface fender derivative amp with american speakers. Ergo not much by the way of mids to start off with..
