If I have a guitar set up to play in standard tuning one step down (D standard), is it okay to drop the low D down another step (Thereby making it drop C) without readjusting the locks themselves?

Its not going to do anything unless you unlock the top before you downtune. If thats what your asking

You don't have to unlock the tuners for tuning, the only locking system like that is if you had a locking nut.
