How does the 250k pot affect the sound a humbucker puts out? Like if I put a hot humbucker through it, will it make it...less hot?

It will darken the tone. The 250k pot will bleed more treble to ground than a 500k pot. The 250k pots were used in strats to tame some of the treble of single coils. If you want less hot just turn the guitar volume down.

So if I were to get a bright humbucker to use through a 500k pot, I should get one slightly brighter to compensate?

No, higher resistance pots let MORE treble through so it'd be fine and dandy...humbuckers usually use 500k pots.
