two questions: 1) How would I go about doing this on a build?
2) What would be the effect on the guitar overall?

Route chambers into the body, and cover it up with a figured wood top.

It may lessen the sustain, it will be much lighter, and it may have a different tone.

In my experience the guitar tends to be more responsive and feels more alive if you put large tone chambers in. If its going to be a heavy rock/metal guitar I wouldn't chamber it very much coz you loose a little bit of the tightness, punch and definition if you take too much wood out.
